- Qualified equipment — production and utility systems that have completed Installation, Operational, and Performance Qualification (IQ/OQ/PQ) and are maintained on a documented calibration/preventive-maintenance schedule.
- An independent quality unit — a quality assurance function organizationally separate from production, with authority to approve or reject batches, investigate deviations, and release or reject incoming materials.
- Documented, followed procedures — written SOPs, executed batch production records, and a deviation/CAPA system, all subject to the data-integrity expectations FDA articulates through the ALCOA+ framework.
- Trained, gowned, monitored personnel — documented training on the specific procedures each person performs, plus environmental and personnel monitoring in classified areas.
- Process validation — evidence, per FDA’s Process Validation lifecycle guidance, that the manufacturing process reliably produces conforming product across the three stages of design, qualification, and continued verification.
Regulatory basis
The core US drug cGMP regulations are 21 CFR Parts 210 and 211, which set the minimum requirements for the methods, facilities, and controls used in manufacturing, processing, packing, and holding a finished pharmaceutical. Biologics manufacturers layer Part 211 requirements together with the biologics-specific provisions in 21 CFR Parts 600-680. Medical device manufacturers work under 21 CFR Part 820 (the Quality System Regulation), which FDA has been transitioning toward alignment with ISO 13485 under the Quality Management System Regulation (QMSR). API manufacturers commonly reference ICH Q7, the internationally harmonized Good Manufacturing Practice Guide for Active Pharmaceutical Ingredients, which FDA and other major regulators recognize as the GMP standard for that segment of the supply chain.
“cGMP facility” vs. “cGMP-certified facility”
Searchers often look for a “cGMP-certified facility,” but FDA does not issue GMP certificates the way an ISO certification body issues an ISO 9001 or ISO 13485 certificate. In the US system, a drug-manufacturing site registers and lists with FDA under the agency’s drug establishment registration and listing requirements, and FDA verifies ongoing cGMP compliance through periodic and pre-approval inspections rather than through a one-time certification event. An inspection results in a classification — commonly summarized as No Action Indicated, Voluntary Action Indicated, or Official Action Indicated — and significant findings are documented on a Form FDA-483 and, if unresolved, can escalate to a Warning Letter. “cGMP-certified” in vendor and marketing material generally means the site has a track record of passing FDA (and often other regulators’, e.g. EMA or MHRA) inspections without significant findings, not that a certificate was awarded. Some jurisdictions outside the US do issue formal GMP certificates through their national authorities; that is a separate, non-FDA process and should not be assumed to apply to a US-based site.
Phase-appropriate GMP: not every facility needs full Part 211
Full cGMP facility status is not required at every stage of drug development. Under 21 CFR 210.2(c) and FDA’s guidance “CGMP for Phase 1 Investigational Drugs” (2008), most Phase 1 investigational drug material is exempt from the full scope of Part 211 and instead follows a reduced, phase-appropriate set of controls. That exemption ends once a drug enters Phase 2 or Phase 3 trials, or once material manufactured under the exemption is used in a later-phase or marketed product — at which point the manufacturing site must meet full Part 211 requirements. This is why an academic or early-stage biotech production suite that is adequate for first-in-human material is often not, by itself, a cGMP facility in the full sense the term implies for a commercial or late-phase manufacturing site.
