Research Animal

An animal that is used, or intended for use, in research, research training, testing, or experimentation, and that therefore falls under one or both of two overlapping US regulatory frameworks: the Animal Welfare Act (which reaches warm-blooded species other than purpose-bred rats, mice, and birds) and PHS Policy on Humane Care and Use of Laboratory Animals (which reaches any live vertebrate used in PHS/NIH-funded work, including species the AWA excludes). Whether a specific animal is a "research animal" for compliance purposes depends on which framework applies to it and to the funding/registration status of the institution using it.

Editorial commentary

A research animal is any live animal used, or intended for use, in biomedical, behavioral, or agricultural research, testing, or teaching. In US regulatory practice there is no single legal definition of the term — whether a given animal counts as a "research animal" for compliance purposes depends on which of two overlapping federal frameworks applies, and the two frameworks define "animal" differently. This entry orients between those frameworks; for the committee that reviews and approves the work itself, see IACUC, and for the underlying statute, see Animal Welfare Act (AWA).

Two overlapping regulatory definitions

Whether an animal is regulated, and by what mechanism, depends on which of two legal bases reaches it:

  • Animal Welfare Act (AWA), 7 U.S.C. §§ 2131–2159, enforced by USDA/APHIS via the Animal Welfare Regulations at 9 CFR Chapter I, Subchapter A. AWA defines "animal" (7 U.S.C. § 2132(g)) as any live or dead dog, cat, non-human primate, guinea pig, hamster, rabbit, or other warm-blooded animal used in research, teaching, testing, experimentation, or exhibition. It explicitly excludes purpose-bred rats of the genus Rattus and mice of the genus Mus, purpose-bred birds, horses not used in research, and farm animals used for food/fiber production. Cold-blooded species (fish, amphibians, reptiles) and invertebrates fall outside the AWA definition entirely, since the statute only reaches warm-blooded animals.
  • PHS Policy on Humane Care and Use of Laboratory Animals, administered by NIH’s Office of Laboratory Animal Welfare (OLAW) under the Health Research Extension Act of 1985. PHS Policy applies to any live vertebrate animal used in PHS-supported (including NIH-funded) research, research training, or testing — a materially broader definition than the AWA’s, because it reaches purpose-bred mice, rats, and birds that the AWA excludes by name.

The practical effect: an institution that receives PHS/NIH funding and also holds USDA research-facility registration typically routes essentially all vertebrate animal use through IACUC review regardless of which statute technically requires it for a given species, often by adopting the Guide for the Care and Use of Laboratory Animals as institutional policy. A purpose-bred laboratory mouse in an NIH-funded study, for example, is a "research animal" under PHS Policy even though it is not one under the AWA; a pet-store rabbit acquired for the same study is a research animal under both.

What falls outside both definitions

Species that are neither warm-blooded (excluding them from the AWA) nor vertebrate (excluding them from PHS Policy) sit outside federal animal-welfare oversight entirely, even though they are genuinely used in research. Drosophila melanogaster (the fruit fly), Caenorhabditis elegans, and most other invertebrate model organisms are the clearest counter-example: widely used research subjects that are not "research animals" in the regulatory sense discussed here, so no IACUC protocol or AWA registration attaches to their use. Zebrafish, by contrast, are vertebrates and so fall under PHS Policy (and typically institutional IACUC oversight) even though they are cold-blooded and therefore outside the AWA.

The 3Rs principle

The 3Rs — Replacement, Reduction, and Refinement — is the foundational ethical framework for animal research, first articulated by William Russell and Rex Burch in their 1959 book The Principles of Humane Experimental Technique:

  • Replacement — substituting non-animal or non-sentient methods (in vitro models, computational simulation, lower-order organisms) for conscious, higher-order animals wherever a scientifically valid alternative exists.
  • Reduction — using the minimum number of animals consistent with obtaining scientifically valid, statistically sound results.
  • Refinement — modifying procedures, housing, and care to minimize pain, distress, and suffering, and to improve welfare, for animals whose use cannot yet be avoided.

The 3Rs are not merely aspirational. The Guide for the Care and Use of Laboratory Animals (8th edition) frames them as a core concept underlying humane animal-use program design, and both AWA-mandated and PHS Policy-mandated IACUC protocol review require investigators to describe the species and approximate number of animals to be used, justify that number, and describe consideration of alternatives to procedures likely to cause pain or distress — the 3Rs, applied at the protocol level. USDA’s National Agricultural Library (Animal Welfare Information Center) and NIH OLAW jointly co-host an annual 3Rs symposium reflecting the framework’s shared standing across both regulatory tracks.
